- 다양한 질병 조건에서 장내 미생물오타의
치료 도구로서의 의의와 응용을 평가하는 증거가
증가하고 있다. 미생물오타는 또한 인간 환자들에게
있어 장내 면역의 중요한 변조자로 묘사되어 왔다.
그러나 진화적으로 인간의 숙주와 공존하는 미생물
종의 수를 고려할 때 면역 조절 활동과 관련된 것은
극히 일부 미생물 종뿐이다. 또한, 알려진 효과자
분자와 특정 작용 메커니즘이 없는 경우 임상
치료제로서의 그러한 퐁코마이티콘'의 중요성이
저하된다. 여기서 나는 규제 t 세포를 억제하면서
세포독성 t 세포 활동을 강화시키는 세균성 균주
Lactobacillus plantarum IMB19(Lp IMB19)를
확인했다. (LpIMB19) 강화된 CD8+ T 세포를 가진

생쥐의 대장화(Colonization) 흑색종에서 매개된 종양
내성. 또한, 이러한 박테리아로부터 특별히 격리되어
CD8 T 를 생산하는 인터페론-y 를 크게 자극하는
정제된 캡슐형 다당류(CPS)는 2 개의 선형
반복단위로 구성된 새로운 화합물로, 람노스 가
풍부하고 phospho-diester 교량을 통해 상호 연결된
9개의 단당류로 구성되어 있다. 기계적으로. CPS는
CD8 T 세포의 강화된 프라이밍으로 이어지는 염증성
대식세포 표현형을 증가시킨다. CPS 는 또한 종양
미생물의 대식세포에 의한 철의 격리를 증가시켜
반관절 면역반응을 증가시킨다. 이러한 결과는
정제된 Lp IMB19나 LP IMB19-CPS가 새로운 항암
"onco-microbiotic"으로 발전할 수 있는 큰 잠재력을
가지고 있음을 시사한다.
There is a growing evidence appreciating the significance and
application of gut microbiota as therapeutic tool in various disease
conditions. Intestinal commensal microbiota has also been described as
a significant modulator of anti-tumor immunity in human patients.

However, only a few microbial strains have been associated with immune
modulatory activity given the sheer number of microbial species which
have evolutionarily co-existed with human hosts. In addition,
significance of such ‘oncomicrobiotics’ as clinical therapeutics is
undermined in the absence of a known effector molecule/s and specific
mechanism of action.
Here, I have identified a bacterial strain Lactobacillus
plantarum IMB19 (Lp IMB19) enhancing cytotoxic T cell activity while
suppressing regulatory T cells. Colonization of mice with (Lp IMB19)
enhanced CD8+ T cell -mediated anti-tumor immunity in melanoma.
Further, purified capsular polysaccharide (CPS) isolated from these
bacteria specifically and significantly stimulates interferon- producing
CD8+ T cells.
Structurally, capsular polysaccharide is a novel compound
composed of a linear repeating unit, rich in rhamnose and made of nine
monosaccharides interconnected via a phosphodiester bridge.
Mechanistically, CPS augments inflammatory macrophage phenotype
leading to an enhanced priming of CD8+ T cells. CPS also increases
sequestration of iron by macrophages in tumor microenvironment
boosting the anti-tumor immune response. These results suggest that
purified Lp IMB19 or Lp IMB19-CPS have a great potential to be
developed as novel anti-cancer ‘oncomicrobiotic’.
