- In this thesis, colorimetric detection of antibiotics using the capture based on covalent bonding and catalytic reaction of platinum nanoparticles was introduced to detect antibiotics.
Firstly, chemical binding sandwich assay for antibiotics detection was developed. In conventional methods, most antibiotics detection methods based on immunoassay were competitive assay. This is because antibiotics detection could not be used by using sandwich immunoassay due to its small size. However, competitive assay had a risk of false negative results. For removing that risk, sandwich immunoassay should be developed for accurate information delivery. It was possible to use chemical sandwich assay by using covalent bonding chemistry instead of capture antibodies. Therefore, only one type of antibody was needed to detect the antibiotics without the risk of false positive results.
Secondly, colorimetric detection method was introduced to detect antibiotics by naked eye. In conventional methods, analytical equipment for detecting antibiotics should use which were very expensive and bulky. It was possible to develop easy and cheap colorimetric detection method by using catalytic reaction of platinum nanoparticles. Because of high catalytic effect of Pt nanoparticles, organic molecules such as 3,3',5,5'-tetramethylbenzidine (TMB) could be oxidized effectively.
