Abstract
세포 간 나노튜브(Intercellular nanotube)는 세포와 세포 사이에 형성되는 얇고(수백 나노미터) 긴(최대 백 마이크로미터 이상) 관으로, 2004년 최초로 보고된 이래 다양한 배양세포들과 조직에서 소포체, 핵산, 신호전달 물질, 소기관 등의 수송을 담당하는 것으로 밝혀졌다. 그러나 그 형태 및 기능의 복잡∙이질성으로 나노튜브의 정의조차 합의되지 못한 채 그 생물학적 의미와 기능에 대한 논쟁이 계속되고 있다. 이러한 이면에는 나노튜브를 이해하는 데 있어 필수적인, 구체적인 구조와 형성과정에 대한 연구 및 이해가 결여되었다는 이유가 있다. 본 연구는 생물물리학적 관점에서 나노튜브가 어떠한 구조를 가지고 있으며, 그러한 구조가 어떤 과정을 거쳐 형성되는지, 그리고 그러한 구조를 결정하는 생물학적 요인이 무엇인지 탐구하였다. 또한 그러한 구조와 나노튜브의 수송 기능 사이에 어떠한 관련이 있는지도 밝히려 하였다. 이를 위해 본 연구에서는 먼저 살아있는 세포의 나노튜브를 이미징하는 동시에 그 역학적 성질을 조사하기 위한 방법으로서, 선스캔 공초점 현미경(Line-scanning Confocal Microscopy)와 광학 집게(Optical Tweezers)를 결합한 단일분자 광학이미징-힘분광학 기술(Single-molecule Optical Imaging - Force Spectroscopy)을 개발하였다(4.1 장). 특히, 본 연구의 목적에 부합하도록 공초점 현미경의 심도(Depth of Field)를 빠른 ETL(Electrically Tunable Lens) 스캐닝을 통해 두 배까지 확장할 수 있는 기술을 개발, 적용함으로써 기존 결합기술의 한계를 넘어 3차원 상의 관찰 분자의 움직임을 추적할 수 있도록 하였다. 다른 색의 형광으로 나노튜브를 구성하는 액틴 섬유(F-actin) 또는 세포막(Plasma membrane)을 염색하고 두 세포 사이의 형성되는 나노튜브의 구성을 관찰한 결과, 기존의 인식과는 달리 나노튜브는 이질적인 다양한 구성상태를 가지고 있음을 확인하였다. 특히, 양쪽 세포의 촉수가 끝 쪽에서 붙거나 겹쳐있는 형태(Hetero-NT)가 한 세포의 촉수만이 확장되어 연결되는 형태(Homo-NT)보다 주를 이루었다. 이어진 나노튜브 형성과정의 실시간 동역학 연구에서 나노튜브들은 최초 형성 단계에서 오로지 Hetero-NT, 특히 끝 쪽에서 연결되는 형태(end-connected)로 형성됨을 관측하였다. 또한 이렇게 형성된 Hetero-NT는 겹쳐있는 형태로 발전한 이후 선택적으로 Homo-NT로 변화하는 것을 확인하여, 앞서 관찰한 다양한 구성상태들 사이의 형성 단계를 규명하였다(4.2 장). 광학 집게를 이용, 나노튜브에 물리적인 외력을 가하는 실험에서 나노튜브는 기존의 보고와 달리 역학적으로 안정한 구조를 가지고 있음을 확인하였다. 또한 외력에 대한 반응 양상을 통해 나노튜브를 세 종류로 분류할 수 있었는데, 각 종류는 앞서 발견한 구성상태와 밀접한 관련이 있었다. 특히 최초 형성 단계라 여겨지는 끝 쪽이 연결된 형태와 기타 형태 사이에는 역학적으로 확연한 차이가 있어, 나노튜브의 형성 과정에서 이러한 강도의 증가를 야기하는 단계가 존재할 것으로 예측되었다(4.3 장). 마이오신 모터 단백질(Myosin)은 촉수의 운동을 조절하는 역할을 하는데, 나노튜브에서도 마찬가지로 선형적인 힘(Myosin-II)과 토크(Myosin-V)를 작용하며 이들의 균형으로 나노튜브의 구조가 형성 됨을, 약물처리 실험에서 확인하였다(4.4 장). 그러나 이들이 작용하는 힘과 토크에서 예측되는 복잡한 미세구조는 기존의 공초점 현미경의 해상도에서는 확인할 수 없었으므로 초고해상도 현미경인 dSTORM 기술로 나노튜브를 관찰하였으며, 그 결과 겹쳐있는 구조의 나노튜브들은 양쪽 촉수가 나선형으로 꼬여있는 형태를 갖고 있음을 최초로 규명할 수 있었다. 마찬가지로 약물처리를 하였을 때 이러한 꼬인 형태에 변화가 일어나, Myosin-V 단백질에 의한 토크가 이러한 꼬인 구조를 야기함을 확인하였다. 또한 이러한 꼬인 구조가 나노튜브의 형성 과정에서 강도를 증가시키는 원인일 수 있음을 고찰하였다(4.5 장). EGFR 단백질은 세포막에 존재하는 신호전달단백질로, 촉수에서 액틴 섬유의 움직임을 따라 세포 안쪽으로 수송됨이 보고된 바 있다. 본 연구에서는 나노튜브에서 이러한 액틴 섬유에 의한 EGFR 단백질의 단방향 수송이 마찬가지로 존재함을 관찰하였다(4.6 장). 나노튜브 구조 및 형성 과정에 대한 본 생물물리학 연구는 그간 결여되었던 나노튜브에 대한 구조적∙역학적 이해를 제공하고 나노튜브의 형성 단계에 대한 모델을 정립하였다는 점에 의의가 있다.
Extensive studies on intercellular nanotubes (NTs) that are formed by a physical connection between protrusions from cell plasma have revealed their biological significances as a long-range pathway for cellular component transport and signal exchange. However, it remains mysterious how a fine structure is formed and sustained robust over long distance for several hours. To address the mechanism of NT formation, we used single-molecule biophysical approaches that can visualize the process of real-time formation of NTs in living cells. Various methodologies of single-molecule biophysics enable us to observe real-time dynamics (~ submicrometers), force (~ piconewtons), and detailed nanostructure (~ nanometers) of biomolecular target. To obtain both seemingly processes of formation and invisible mechanical change in live cell simultaneously, we developed the novel approach of advanced real-time single-molecule optical imaging-force spectroscopy. To obtain an optical signal outside the focal plane without unintended interruption of the force signal in this imaging system, we developed an optical method to extend the depth of field in a high numerical aperture objective ( 1.2), required to visualize a single fluorophore. Our approach is simple, fast and inexpensive, but it is powerful for imaging target molecules axially in single-molecule optical imaging-force spectroscopy, especially for NT study in live cell. As a result, we could track both NT structure by fluorescent imaging and related mechanics by optical tweezers. We observed that NTs between two differently labeled cells have heterogeneous configurations in either F-actin or membrane labeled cells. NTs were initially formed by a direct contact between filopodia protruded from the plasma membrane of cells and evolved dynamically into a long, single filopodial track (Homo-NT) through intermediate overlapped configuration (Hetero-NT). By manipulating physically NTs, we found that NT has highly robust structure resisting to external force up to 60 pN without any breaking. We classified distinct mechanistic classes of NTs with different robustness and found that each class corresponds to different states of NT biogenesis while the critical mechanical change exists for a specific stage. The change of an initial flexible NT to a stiff NT can be induced by inner chemical changes of NTs, or possibly, by the mechanical and structural change, which has not been addressed. We examined that molecular motor proteins, non-muscle myosin-II and myosin-V, apply retracting force and torque, which cause the conformational change of NT. Consequently a double helical structure of Hetero-NTs induced by the torque was identified by super resolution microscopy (dSTORM), which is an unreported structure of NTs that have been recognized only as a linear structure. This unique and elaborate structure is a convincing substitute that results in the structural change of stiffening NTs. Moreover, by axial scanning, using an electrically tunable lens with a fixed sample, we were successfully able to visualize the epidermal growth factor receptor (EGFR) moving along the NT three-dimensionally while another EGFR on the intercellular nanotube was trapped by optical tweezers in living cells. This result proves that NTs plays a role of a bridge between cells for the transport of activated receptor proteins. Our intensive biophysical studies on NTs reveals mechanistic NT biogenesis of cultured cell in a nanometer-piconewton scale, from initially flexible to stiff states, related EGFR transport and open the window to an abundance of new research directions of the profound studies on NT formation.
