Abstract
In this study, an electrolyte-gate field effect transistor (EGT) device was manufactured for the purpose of detecting target molecules contained in liquid samples. EGTs have advantages of label-free, low cost, high sensitivity, and low power. EGT with silicon nanonet (SiNN) channel was designed to secure high current-related sensitivity with the same voltage-related sensitivity due to its excellent electrical characteristics. Conventionally, chemical surface treatment, receptor immobilization, and receptor-target binding were performed on the channel part of the EGT as a sensing layer, but the probability of binding is low due to small nanoscale area of the channel. Therefore, a study was conducted to improve the sensitivity by using the gate electrode having a larger area on a microscale as a sensing layer. Based on these advantages, SiNN EGT was utilized for peanut allergen (PA) detection, and highly sensitive and selective detection were demonstrated. Furthermore, the sensitivity was optimized using analytical modeling and verification with PA detection. First, for uniform and sensitive detection of biomolecules, superior electrical characteristics and uniform distribution are emphasized when the EGTs were fabricated. The EGT device was fabricated using a top-down process suitable for industrial production. The SiNN channel was defined using a step-and-scan method with low cost and high throughput instead of conventional electron beam lithography. The fabricated SiNN EGT showed excellent intrinsic electrical characteristics, including a low threshold voltage of 0.7 V, low subthreshold swing of <70 mV/dec, and low gate leakage of <10 pA. The applicability of EGT as a sensor was verified with pH test. Second, actual PA detection was carried out using the manufactured SiNN EGT device. PA should be detected rapidly and accurately because it has a fatal effect on the human body such as anaphylactic shock, blindness, and breathing difficulties. The concentration of PA was prepared as 1 pg/mL–1 mg/mL through decimal dilution, and the chemical surface functionalization for receptor immobilization was conducted. The transfer curve shift and sensitivity when reacting with a PA sample were analyzed, and the transfer curve shifted to the left as PA concentration increased. The current-related sensitivity was significantly increased as the operation regime is varied from linear regime to subthreshold regime due to high transconductance at subthreshold regime. The SiNN EGT showed selective detection of PA through a non-specific control test. The limit of detection (LOD) was extracted from the current-related sensitivity, which was 10 times lower LOD than the conventional PA sensor. Finally, a lumped-capacitive model was established for an EGT with two sensing layers of the gate electrode and the channel. Through an analysis of the effect of the receptor-target bindings at each sensing layer on the sensitivity, the operation regime for high sensitivity can be predicted. The voltage-related sensitivity is mainly determined by the effective dipole potential (VDP,EFF), the effective capacitance of the functionalization layer (CFN,EFF), and the total charge density in the channel (QS) related to the operation regime. In an n-type EGT, the sensitivity is enhanced as the device is operated in the subthreshold regime with a positive VDP,EFF and a small CFN,EFF. This is a meaningful technique that can extract the optimal operation regime for various directions, magnitudes, and types of dipole moment and capacitance. The influence of the VDP,EFF and CFN,EFF was experimentally evaluated by detecting the PA using the fabricated n-type SiNN EGTs. As a result, positive VDP,EFF was extracted from the measurement, so subthreshold regime was expected as an optimal operation regime. Actually, the voltage-related sensitivity in the subthreshold region was lower than the linear region, and the LOD in the subthreshold region was 70 pg/mL, which was twice lower than the linear region. Therefore, the model suggests a method to improve and optimize detection performance in terms of sensitivity and LOD.
본 연구에서는 액상 상태의 시료에 포함된 표적물질 검출을 목적으로 전해질게이트 전계 효과 트랜지스터 소자를 제작하였고, 이를 기반으로 바이오물질 검출을 수행 및 분석하였다. 산업, 환경, 의료, 군사 등의 실시간, 고감도의 바이오물질 검출이 필요한 분야가 점점 광범위 해지며, 검출의 중요성이 강조됨에 따라 독창적인 바이오센서 개발의 필요성이 부각되고 있다. 다양한 바이오센서 플랫폼 중, 전해질 게이트 전계 효과 트랜지스터를 이용한 바이오센서가 높은 관심을 받고 있다. 표지자가 필요 없는 검출 방식 임에 따라 실시간 검출이 가능하며, 반도체 소자를 제작하는 표준 공정 방식인 CMOS 공정 방식과 호환이 됨에 따라 저비용 공정이 가능하다. 또한, 바이오물질과 비슷한 크기의 감지부 영역을 이용함에 따라 고민감도, 수용체-표적물질의 결합에 의한 전기신호를 검출함에 따라 고선택도의 특징을 가진다. 이번 연구에서는 위와 같은 장점을 가지는 전해질 게이트 전계 효과 트랜지스터를 기반으로, 실리콘 나노넷 구조의 채널을 이용하여 우수한 전기적 특성을 확보함에 따라 동일한 전압민감도 대비 높은 전류민감도를 확보할 수 있도록 소자를 디자인하였다. 더불어, 기존에는 소자의 채널부를 감지부로 이용하여 화학적 표면처리, 수용체 고정 및 수용체-표적물질 결합을 진행하였지만 이는 나노스케일의 작은 면적에서 진행되는 문제점이 있어 결합의 확률이 적다는 단점이 있었다. 따라서, 본 연구에서는 마이크로스케일의 더욱 큰 면적을 가지는 게이트 전극을 채널과 더불어 감지부로 이용하여 민감도를 향상시키기 위한 연구를 수행하였다. 산업적 생산에 적합한 탑다운 공정 방식을 적용하여 실리콘 나노넷 구조의 전해질 게이트 전계 효과 트랜지스터 소자를 제작하였다. 실리콘 나노넷 채널은 기존의 전자빔 리소그래피를 대신하여, 비용이 적으며 전사 속도가 빠른 step-and-scan 방식을 이용하여 제작하였다. 전기적 특성 평가 결과, 문턱전압 ~0.7 V, 문턱전압이하스윙 ~70 mV/dec, on/off 전류비 ~107, 게이트 누설전류 <10 pA 로 우수한 전기적 특성을 확보하였으며, pH 민감도 측정을 통해 센서로서의 활용 가능성을 검증하였다. 제작된 소자를 이용하여, 실제 땅콩 알러젠 검출을 진행하였다. 아낙필락시스 쇼크, 실명, 호흡 곤란 등 인체에 치명적인 영향을 주는 물질인 땅콩 알러젠은 신속 정확한 검출이 요구되는 바이오 물질이다. 검출에 앞서 땅콩 알러젠의 농도는 10진 희석을 통하여 1 pg/mL–1 mg/mL 로 설정하였다. 땅콩 알러젠 시료와 반응 시 변화하는 특성 곡선 및 민감도를 분석하였으며, 고농도의 땅콩 알러젠과 반응할수록 특성 곡선은 왼쪽으로 이동하였다. 또한, subthreshold 영역에서의 전류민감도가 linear 영역보다 높았으며, 이는 동일한 게이트 전압 변화에 대해 subthreshold 영역에서의 전류 변화가 지수적으로 증가하기 때문이다. 선택도 검증 실험을 통하여, 표면처리된 수용체는 선택적으로 땅콩 알러젠을 검출할 수 있을 또한 검증하였다. 3-sigma 방법을 이용하여 추출된 검출한계는 기존의 땅콩 알러젠 센서에 비해 10배 적은 결과를 확보할 수 있었다. 나아가, 게이트 전극과 채널 두 영역을 감지부로 이용하는 전해질 게이트 전계효과 트랜지스터에 대한 모델링을 수행하였다. 두 감지부에서의 반응이 민감도에 미치는 영향을 분석함에 따라, 높은 민감도 확보가 가능한 동작 영역을 예측할 수 있으며 실증실험을 통하여 확인하였다. 수용체-표적물질의 반응은 dipole moment 와 capacitance 두 전기적 요소의 형태로 표현이 가능하며, 문턱전압, 문턱전압이하스윙 등의 소자의 전기적 특성이 변화하며 이에 따라 동작 영역에 따라 민감도가 달라진다. 이는 dipole moment 와 capacitance 의 방향성, 크기 및 종류에 따라 최적화된 동작 영역을 추출할 수 있는 기법을 제시하는데 의의가 있다. 실제 땅콩 알러젠 시료를 이용한 검출 결과, subthreshold 영역에서의 전압 민감도가 linear 영역에서 낮았으며, subthreshold 영역에서의 검출한계는 70 pg/mL 로 linear 영역에 비해 2배 낮은 수준의 결과를 확보하였다. 해당 모델은 민감도와 검출한계 측면에서 검출 성능을 향상 및 최적화하는 방법을 제시한다.
