Abstract
Tau protein 은 신경 세포를 고정하는 단백질로 Tau protein 이 세포에서 빠져나와 자기들끼리 결합하는 것을 tauopathy 라 하며, 이로 인해 세포 내 Tau protein 이 부족해지면 신경 세포와 뇌에 손상이 발생하며, 연구자들은 이를 알츠하이머병의 원인 중 하나로 보고 있고 알츠하이머병의 원인 해석에 Tau Protein 의 정량적인 분석이 중요하다. 이미 상용화되어 있는 ELISA 이외에도 Sandwich Antibody Microarray 등이 있지만 본 연구실에서 개발한 AFM 방법을 활용하면 혈액 내 낮은 농도의 Tau Protein의 분석도 가능할 수 있다. 이에 대한 기반 연구로서, 가장 size가 큰 Tau Protein인 Tau-441과 해당 protein에 epitope를 갖는 antibody 간의 상호작용을 확인하기 위해 AFM 을 이용한 force spectroscopy 에서 힘과 rupture distance 등의 정보를 얻어 protein 을 검출하고 나아가 정량적인 분석에 있어서 어떤 민감도를 나타내는 지 확인하기 위하여 여러 농도의 Tau Protein 에 대한 실험을 수행하였다.
Tau protein is a protein fixing nerve cells. Tauopathy is autopolymerization of tau protein, resulting in depletion of the protein in cells. It is believed that such low concentration makes damage to nerve cells and Brains, and it is associated with Alzheimer’s disease. Therefore, it is important to analyze Tau protein in blood or cerebrospinal fluid quantitatively to understand Alzheimer’s disease. Typically, either sandwich antibody microarray or ELISA is employed for the analysis, but AFM approach developed in this laboratory suggests that it may be possible to analyze Tau protein of biologically relevant concentration. In this study, various concentrations of Tau-441 which is the largest one among tau protein family were examined, and the rupture distance and force value were obtained to confirm the specific antigen-antibody reaction of tau protein. Whereas detection limit of the current approach is close to that of the conventional methods, it is believed that the limit can be enhanced, if a smaller spot is employed for the analysis.
