Abstract
나는 박사학위 논문을 위해 두 가지 주제의 프로젝트를 진행했고, 이 과정은 비만과 종양 연구 분야에 대한 새로운 개념을 이해하고 발전시키는데 기여했다. 내 논문의 첫 번째 주제는 '비만을 조절하는데 있어 대식세포의 저산소 유도 인자 1(Hypoxia Inducible Factor-1; HIF-1)의 역할'이다. 비만에서 지방세포는 크기가 커지면서 조직의 저산소증을 유도하게 되고 대식세포들을 지방조직으로 모이게 한다. 저산소 조건 하에서 활성화되는 전사인자 HIF의 지방세포에서의 역할은 이전 연구들을 통해 많이 보고되었지만, 지방조직 대식세포의 HIF가 비만에 어떤 영향을 미칠 수 있는지는 아직 잘 밝혀지지 않았다. 나는 myeloid 세포 특이적인 Hif-α 결여 마우스에게 고지방 식이를 먹였고, 이 Hif-1α 결여 마우스가 상대적으로 살이 찌지 않는 것을 확인하였다. 나는 또한 myeloid 세포 특이적인 Hif-1α 결여 마우스가 지방분해 능력이 높고, 열발생이 증가되어 있다는 것을 관찰했다. 이들 마우스부터 분리된 지방조직 대식세포는 M2 대식세포의 특징을 가졌으며 미토콘드리아 기능이 증가되어 있었고, uncoupling protein 1 (Ucp1) 발현량이 증가되어 있었다. 또한, 이 지방조식 대식세포는 열을 발생시켜 지방 조직의 지방분해를 조절했다. 또한, 대식세포는 acetyl-CoA의 공급원인 acetate 처리로 열을 생산할 수 있었지만 Ucp1 발현이 감소했을 때는 열을 생산하지 못하였다. 이는 지방조직 대식세포의 열 생성에 Ucp1의 발현이 꼭 필요하다는 것을 보여주는 결과이다. 마지막으로, 나는 마른 마우스와 마른 환자들의 지방 조직에서 UCP1을 발현하는 대식세포가 증가 되어있다는 것을 발견했다. 요약하자면, 지방조직 대식세포에서 UCP1에 의한 열 발생이 지방조직의 지방분해를 증가시켜 다이어트에 의한 비만을 예방한다는 것을 보여준다. 내 논문의 두 번째 주제는 ‘초고선량 FLASH 방사선이 Lewis lung carcinoma의 종양 미세환경에 미치는 영향’이다. 초고선량 FLASH 방사선 조사는 최근 다양한 동물 모델에서 폐, 내장, 뇌, 피부 등의 정상 조직의 손상을 줄이고 효과적인 항암능력을 가진다고 알려졌다. 하지만 이러한 정상 조직의 손상을 줄이는 메커니즘에 대해서는 아직 알려진 바가 없다. 나는 이 연구에서 마우스의 Lewis lung carcinoma를 이용하여 기존 방사선 조사와 FLASH 방사선 조사가 종양 미세환경에 영향을 미치는 다른 효과를 밝혀냈다. 기존 방사선 조사는 FLASH 방사선 조사와는 달리 혈관 수축의 증가, phosphorylated myosin light chain (p-MLC) 의 활성화, 그리고 종양내 γH2AX 발현 세포의 증가를 유도한다는 것을 면역염색법을 통해 찾아냈다. 또한, 나는 MLC kinase의 억제제인 ML-7이 Lewis lung carcinoma에서 방사선 유도 γH2AX 생성을 감소시키는 것을 확인했다. 마지막으로, 나는 마우스에 기존 방사선 조사를 하고 동시에 ML-7을 처리하여 혈관 수축 감소, γH2AX 발현 세포 감소, CD8α 양성 T세포와 S100A8 양성 골수세포의 침투 증가 등 FLASH 방사선 조사와 유사한 효과를 보았다. 요약하자면, 나는 이 논문을 통해 기존 방사선 조사와 플래시 방사선 조사 사이에서 차별적으로 제어되는 MLC 활성화를 조절하는 것이 방사선 치료를 개선하는 새로운 타겟이 될 수 있음을 밝혔다.
I worked on two topics for my Ph.D. thesis and these progresses may contribute to the advances on new concept and understanding in obesity and tumor research area. First topic of my thesis is ‘The role of Hypoxia-Inducible Factor-1 (HIF-1) in macrophages in regulating obesity’. During obesity, adipocytes expand creating tissue hypoxia and recruit macrophages into the adipose tissues. Although previous studies have reported the role of HIF, a transcription factor activated under hypoxic conditions in adipocytes in obesity, it is poorly understood how HIF in adipose tissue macrophages (ATM) could impact obesity. Here I challenged myeloid-specific Hif-α knockout (KO) mice to high fat diet and observed that Hif-1α but not Hif-2 α KO mice were significantly protected from obesity. I also found that theses myeloid-specific Hif-1α KO mice had increased lipolytic activity and improved thermogenesis. ATM isolated from these mice exhibited an increased expression of uncoupling protein 1 (Ucp1) with M2-polarized character and increased mitochondrial functions. These ATM also produced heat, which regulates lipolysis in adipose tissue. However, ATM generated lower amount of heat when Ucp1 expression was decreased although acetate treatment, source of acetyl-CoA, was able to produce heat, indicating Ucp1 was required to produce the heat. Lastly, I found that lean mice and patients had increased UCP1 expression in macrophages in their adipose tissues. In summary, I demonstrated that heat production and UCP1 expression in ATM increase lipolysis in adipocytes thereby preventing the diet-induced obesity. Second topic of my thesis is ‘Effects of ultra-high dose rate FLASH irradiation on the tumor microenvironment in Lewis lung carcinoma’. The ultra-high dose rate FLASH irradiation (IR) has recently been described superior protection effect in normal tissues such as the lung, gut, brain and skin and effective anti-tumor ability in various animal models. However, the mechanism of this protection effect is still unknown. Here I report different effects of conventional dose rate (CONV) versus FLASH IR for producing unique changes in the tumor microenvironment using Lewis lung carcinoma in mice. I found that CONV IR resulted in vessel constriction, increased phosphorylated myosin light chain (p-MLC) level, and much higher numbers of γH2AX (DNA double strand break marker)-positive cells in tumors, which were not observed with FLASH IR by immunostaining. I also demonstrated that ML-7, an inhibitor of MLC kinase decreased IR-induced γH2AX formation in Lewis lung carcinoma in vitro. Lastly, I observed that CONV IR showed similar effects to FLASH IR when ML-7 was treated to mice, which include the decreased vessel constriction, fewer γH2AX expressing cells, and increased CD8α-positive T cells and S100A8-positive myeloid cells. In summary, I believe that regulating of MLC activation which is differentially controlled between CONV and FLASH IR is a novel target.
